ABSTRACT

The spreading of knowledge depends on the access to the information and its immediate use. Models are useful to explain specific phenomena. The scientific community accepts some models in Biology after a period of time, once it has evidence to support it. The model of the structure and function of the DNA proposed by Watson & Crick (1953) was not the exception, since a few years later the DNA model was finally accepted. In Costa Rica, DNA function was first mentioned in 1970, in the magazine Biologia Tropical (Tropical Biology Magazine), more than 15 years after its first publication in a scientific journal. An opposite situation occurs with technical innovations. If the efficiency of a new scientific technique is proved in a compelling way, then the acceptance by the community comes swiftly. This was the case of the polymerase chain reaction, or PCR. The first PCR machine in Costa Rica arrived in 1991, only three years after its publication.

ABSTRACT

Realizou-se um estudo transversal de diagnóstico para validaçäo da reaçäo em cadeia da polimerase (polymerase chain reaction) - PCR no diagnóstico da leishmaniose tegumentar americana - LTA através da determinaçäo de sua sensibilidade, especificidade, valor preditivo positivo e valor preditivo negativo. A amostragem compreendeu 63 pacientes portadores de LTA e 68 sadios. Para inclusäo no primeiro grupo, os pacientes deveriam apresentar pesquisa direta positiva em material obtido por biópsia da borda da lesäo cutânea. No segundo grupo, os critérios de seleçäo incluiam a ausência de lesöes cutâneas sugestivas de LTA e negatividade das reaçöes de Montenegro e imunofluorescência indireta. Os tecidos provenientes das biópsias (borda de úlcera no primeiro grupo; pele normal no segundo) eram colocados em tubos plásticos com tampa e imediatamente congelados (-20 ºC). Cada tubo recebia um número, cuja correspondência com o grupo sadio ou controle era de conhecimento exclusivo do autor. Assim, os profissionais do Centro de Pesquisas René Rachou/Fiocruz envolvidos nas etapas seguintes (extraçäo de DNA e PCR) näo tinham ciência de tal correspondência, garantindo o caráter cego do trabalho. Obteve-se uma sensibilidade de 98,41 por cento e uma especificidade de 95,59 por cento...
